Effect of neem leaf extract on the degradation of IκBα and translocation of p65 and p50 to the nucleus. Jurkat cells were incubated with neem leaf extract (at 300 μg/ml) for 2 h, followed by a TNF-α (20 ng/ml) treatment for the indicated time periods. Cytoplasmic and nuclear extracts were prepared, fractionated on a 10% SDS–page gel, transferred to a membrane, and then tested for IκBα, p50, p65 (cytosolic) or p50 and p65 (nuclear). Equality of protein loading and purity of nuclear/cytosolic extracts was checked by lamin B and α-tubulin Western blots. The data shown here were representative for three independent experiments with similar results
