Prolactin stimulates the movement of ZnT2 from a peri-nuclear localization to an intracellularly dispersed compartment and Zn secretion in normal breast (HC11) cells. Cells were left untreated (a) or treated with prolactin (b, 1 μg/mL) for 30 min then fixed and permeabilized with Triton X-100. ZnT2 was detected with affinity purified ZnT2 antibody and visualized with anti-rabbit IgG conjugated to Alexa 488 (green). Nuclei were stained with TOPRO-3 (red). Images were collected at ×100 under oil. c Treatment of HC11 cells with prolactin (1 μg/mL) that had been pre-loaded with 65Zn stimulated 65Zn secretion.
